How to Inject Semaglutide

Step-by-step instructions, best injection sites, and practical tips to help you inject semaglutide safely and confidently. This guide is designed for patients using semaglutide as part of a clinician-supervised medical weight loss program.

Before You Start

Important: Always follow the dosing plan provided by your licensed medical provider. This guide is for general educational purposes and does not replace individualized medical advice. If you have questions about your dose, timing, or side effects, contact your care team.

Supplies & Preparation

Before injecting, gather the following:

  • Semaglutide vial or prefilled syringe (as prescribed)
  • New insulin syringe or pen needle
  • Alcohol swab
  • Sharps container

Preparation steps:

  1. Wash your hands thoroughly with soap and water.
  2. Clean your chosen injection site with an alcohol swab and allow it to dry.
  3. Inspect the medication. Do not use it if the solution is cloudy, discolored, or contains particles.

Drawing the Medication (Vial Use)

If your medication is provided in a vial:

  1. Remove the vial cap and clean the rubber stopper with an alcohol swab.
  2. Pull back the syringe plunger to the prescribed dose.
  3. Insert the needle into the vial and inject air.
  4. Turn the vial upside down and slowly pull the plunger to the correct dose.
  5. Tap out air bubbles and recheck the dose.

How to Inject Semaglutide

  1. Pinch the skin at the injection site.
  2. Insert the needle at a 90-degree angle.
  3. Inject the medication slowly and steadily.
  4. Remove the needle and release the skin.
  5. Dispose of the needle immediately in a sharps container.

Semaglutide is injected once weekly, on the same day each week, unless otherwise directed by your provider.

Best Injection Sites

Rotate injection sites weekly to reduce irritation and improve absorption.

Common injection areas:

  • Abdomen: At least two inches away from the belly button
  • Thigh: Front or outer thigh
  • Upper arm: Back of the arm (often requires assistance)

Rotation tip: Avoid injecting into the exact same spot two weeks in a row.

Things to Avoid

Avoid injecting into the same spot repeatedly — this can cause irritation or lumps.

Avoid poor hygiene — always clean the skin and use a new needle.

Avoid doubling doses — never inject extra medication if you miss a dose without medical guidance.

Avoid injecting into irritated or bruised skin.
